my boss has just put some more memory in the G4. When opening logic the audio file is present in the arrange window but does not play back. Will playback in audio window. Any ideas why this would happen?

Yes, quite easily. Or slightly incorrect specs for the computer. Get it replaced and see if everything works right.
You could also try to test the memory using:
Don't use the hardware test CD that came with the Mac, it's less then worthless and would only alert you if something was physically on fire. Maybe.
