Out of ignorance, I ask anyone:

How does one boot up in DOS--not running MS-DOS through Windows--as I am upgrading my BIOS and this AFUDOS application (run off a floppy, which I've made, set first boot device to the floppy, etc... anything off the ASUS support site) doesn't seem to start. The message received when I try:

Remove any discs or media.
Press any key to restart.

Another thing:
I should have the .ROM of the new BIOS version on my floppy, not just the executable AFUDOS... right?

Alright, I put in my own logic.

I made an MS-DOS bootup floppy and added the AFUDOS file onto it, along with the .rom. It booted, and I could run the app, but it couldn't read the ROM. Odd.
