Google will not bother about signature as long as the comments are genuine and not spammy, But if you spam then google may penalize your site.

If you are targeting organic traffic then you need to do proper SEO submissions like Bookmarking , Forum , Blog commenting on niche sites are important.

From how many days you are doing off page activities? are you checking the approvals? you need to submit your site to search engines.

It will be accepted once in a bluemoon but it completely depends on the admin, edu sites are very tough to get. It is tougher to get backlinks from edu sites.
